PCR Efficiency:
Efficiency = (10^(-1/slope) - 1) × 100%

Standard Curve:
Ct = slope × log(concentration) + intercept

Concentration from Ct:
Concentration = 10^((Ct - intercept) / slope)

Ideal Efficiency: 90-110% (slope: -3.1 to -3.6)
Important: For accurate qPCR efficiency calculations, ensure your standard curve has at least 5-6 data points spanning 4-6 orders of magnitude. Ct values should be in the linear range of your assay. Poor efficiency may indicate primer design issues, reaction conditions, or template quality problems.
